Identification and expression analysis of goose type lysozyme in half smooth tongue sole (Cynoglossus semilaevis)

The goose-type lysozyme is one of the three major distinct lysozyme types identified in the animal kingdom including teleosts. In this report, we identified, sequenced, and characterized the goosetype lysozyme gene (CsGLys) from half-smooth tongue sole (Cynoglossus semilaevis). The full-length cDNA of CsGLys is 1191 bp in length from the transcription start site to polyadenylation site, including a 91 bp 50-terminal untranslated region (UTR), a 452 bp 30-terminal UTR and a 648 bp open reading frame (ORF) of encoding a polypeptide with 215 amino acids. The deduced amino acid sequence of CsGLys possesses a Goose Egg White Lysozyme (GEWL) domain with three conserved residues (E91, D104 and D121) essential for catalytic activity. MHC polymorphism and disease-resistance to Edwardsiella tarda in six turbot (Scophthalmus maximus) families

One hundred fry from each of six families were infected with Edwardsiella tarda by intraperitoneal injection. Family mortality ranged from 28.0% to 83.3%. Complete exon 2 and intron 1 sequences of MHC Class II B genes were amplified from five survivor and five non-survivor individuals per family using the clone-sequence method. Thirty-seven sequences from 60 individuals revealed 37 different alleles, 25 of which were unique to this study. The 25 unique alleles belonged to 16 major allele types. Nine alleles were used to examine the association between alleles and resistance/susceptibility to disease. Five alleles were present in an individual, suggesting a minimum of three loci or copies of the turbot MHC Class II B gene. 牙鲆胚胎冷凍損傷的觀察
作 者: 王春花 ,田永勝 摘 要: 為了觀察牙鲆胚胎在冷凍保存過程中的形態(tài)受損情況,將其浸入20%PM(20%丙二醇和20% 甲醇1:l的混合液)中平衡,并用程序化法和玻璃化法對(duì)其冷凍保存2h后解凍,用攝影顯微鏡記錄其在抗凍劑里平衡時(shí)和冷凍保存后的形態(tài)。結(jié)果顯示在平衡過程中胚胎卵膜出現(xiàn)凹陷(稱為溶液損傷),但可以恢復(fù);在冷凍過程出現(xiàn)胞內(nèi)冰損傷,是致命的;用程序化法冷凍保存的尾芽期胚胎卵膜和卵黃完好,胚體邊緣受傷;用玻璃化法保存的尾芽期胚胎,卵膜和卵黃損傷較重,胚體損傷較輕;因此將玻璃化法和程序化法結(jié)合可以達(dá)到揚(yáng)長(zhǎng)避短的效果。 -

牙鲆胚胎玻璃化冷凍及解凍過程中的溫度因子與玻璃化率
作 者: 田永勝,劉本偉,陳松林 摘 要: 為避免魚類胚胎玻璃化冷凍過程中脫玻璃化現(xiàn)象發(fā)生,本文對(duì)冷凍過程中溫度變化及解凍方法進(jìn)行了研究。在室溫13℃下,將含有玻璃化液FVS3的麥管直接投入到一196℃ 的液氮中,麥管內(nèi)溫度達(dá)到一196℃ 時(shí),所用的時(shí)間為15.46±1.38 s,降溫速率為800。C/rain。對(duì)玻璃化麥管在35~45。C水浴中解凍時(shí)間進(jìn)行測(cè)定。結(jié)果顯示,解凍所需時(shí)間在6.17~7.94 S之間,麥管從一196℃升高到13。C的解凍速率為1 500~2 000。C/rain。當(dāng)水浴溫度從37℃逐步升高到72。C時(shí),對(duì)水浴表面蒸氣溫度的變化進(jìn)行了測(cè)定,相應(yīng)水溫下蒸氣溫度從25.47℃升高到38.6。C。在麥管中加入不同濃度的玻璃化液FVS1、FVS2、FVs3和FVS4進(jìn)行玻璃化冷凍,然后將麥管分別在37℃ 水浴中和28.6℃的蒸氣中解凍時(shí)玻璃化率進(jìn)行研究。結(jié)果顯示,在蒸氣中解凍時(shí)的玻璃化率明顯高于水浴中解凍;麥管玻璃化率在水浴中解凍時(shí)不穩(wěn)定,在蒸氣中解凍時(shí)表現(xiàn)穩(wěn)定,玻璃化率隨著玻璃化液濃度的升高有逐漸升高的趨勢(shì)。對(duì)含有牙鲆胚胎的玻璃化冷凍麥管分別在37℃水浴和28.6℃蒸氣中解凍,結(jié)果表明,蒸氣中解凍時(shí)的玻璃化率同樣明顯高于水浴中解凍。 -
顯微注射抗凍劑對(duì)牙鲆(Paralichthys olivaceus)胚胎毒性及冷敏感性的影響
作 者: 姜靜,田永勝,翟介明,陳松林 摘 要: :在魚類胚胎冷凍保存中,由于胚胎體積大、卵膜具多層結(jié)構(gòu),并且通透性差,傳統(tǒng)的平衡法無法使抗凍劑足夠、有效的進(jìn)入胚胎內(nèi)部起到保護(hù)作用。為彌補(bǔ)了普通平衡法的不足,此研究利用顯微注射技術(shù),將抗凍劑直接注射到魚類胚胎卵黃囊內(nèi),提高了抗凍劑對(duì)胚胎的保護(hù)程度。此研究以牙鲆胚胎為試驗(yàn)材料,對(duì)顯微注射抗凍劑種類、抗凍劑的注射劑量、注射濃度、以及抗凍劑注射后胚胎的冷敏感性影響進(jìn)行了篩選。結(jié)果表明:牙鲆胚胎較適宜的顯微注射劑量為750pl;幾種抗凍劑注射牙鲆胚胎后的毒性 大小相對(duì)排列為PVP>蔗糖,DMSO>MeOH>PG,其中注射PG獲得的胚胎成活率和孵化率最高,注射PVP獲得的胚胎成活率和孵化率最低,而PG與MeOH組成混合抗凍劑PM毒性更低獲得的胚胎成活率和孵化率更高。而任何一種抗凍劑均隨著濃度的增加其毒性隨之增加;另外,在冷敏感試驗(yàn)中,顯微 注射6 mol/L的PM的胚胎胚胎,應(yīng)用程序化法處理,以2~C/min的速率由室溫降至一20℃ ,平衡10 min后解凍處理,發(fā)現(xiàn)注射PM的胚胎低溫處理后成活率為(25.07~1.57)%,而6mol/的PM五步平衡法同步處理的對(duì)照組胚胎成活率為(20.88士2.84)% ,說明顯微注射的胚胎冷敏感性一定降低。 -

花鱸冷凍精液受精魚苗與鮮精受精魚苗的同工酶比較分析
作 者: 陳艷翠 ,高天翔 ,陳松林 ,季相山 ,田永勝 ,鄧 寒 摘 要: 采用水平凝膠電泳技術(shù)對(duì)花鱸(Lateolabrax maculatus)冷凍精液受精魚苗與鮮精受精魚苗進(jìn)行了同工酶分析。本實(shí)驗(yàn)選取肌肉和肝臟兩種組織,檢測(cè)了IDHP、PGM、LDH、GPI、MDH、G3PDH、ME、HK、SOD、G6PD和SDH等11種酶,得到13個(gè)位點(diǎn):IDHP 、PGM一1 、PGM.2 、LDH 、GPI-1 、GPI-2 、MDH 、G3PDH 、ME 、HK 、SOD 、C.6PD 和SDH 。結(jié)果顯示在0.99水平,SDH 和PGM一2 位點(diǎn)在兩種受精魚苗中均表現(xiàn)為多態(tài),多態(tài)位點(diǎn)比例均為0.1538;平均預(yù)期雜合度分別為0.0460和0.0407,但未觀測(cè)到雜合子,觀測(cè)雜合度均為0,遺傳偏離指數(shù)均為一1;有效等位基因數(shù)分別為1.0720和1.0850,花鱸冷凍精液受精魚苗與鮮精受精魚苗的遺傳相似度為0.9991,遺傳距離為0.0010 -
額爾齊斯河江鱈精子冷凍保存研究

超低溫冷凍保存對(duì)七帶石斑魚胚胎酶活性的影響
